The titular character and host of Pee-wee's Playhouse.
Miss Yvonne
The most beautiful woman in Puppetland.
- Fluffy Fashion Feathers: Some of her outfits have white feather trim.
- "Happy Holidays" Dress: Wore one in the Christmas special.
- World's Most Beautiful Woman: Miss Yvonne is routinely stated (by Pee-wee and others) to be the most beautiful woman in Puppetland, where the Playhouse is apparently situated.
Captain Carl
Character from the original stage show, played by Phil Hartman.
- Father Neptune
- Put On a Bus: He's only seen in Season One and the original stage show.

- Suspiciously Similar Substitute: For Mrs. Steve.

- Only Sane Woman: At least at the beginning of the series, she is confused by the Playhouse and everything in it and is shown to be a bit freaked out by the strange customs such as screaming at the secret word and playing Connect The Dots inside Magic Screen.
The King of Cartoons
The Playhouse Gang
Group of kids who randomly show up to hang with Pee Wee.
- Suspiciously Similar Substitute: The three kids from the first season were replaced by three other kids in the second.
