Fanboy and Chum Chum
- Voiced by: David Hornsby (Fanboy), Nika Futterman (Chum Chum)
The eponymous, hyperactive duo, who live by the rule of no work and all play. They spend most of their free time hanging out in the local convenience store or having all kinds of wacky adventures in their house, which lacks any kind of adult resident. They are both comic book nerds (although much less so than Oz) and get ecstatic about the tiniest things.
- Cloudcuckoolanders
- Cool Mask: Fanboy has one, although he lacks any real powers.
- Cross-Dressing Voices: Nika Futterman as Chum Chum.
- Deuteragonist: Chum Chum.
- Expressive Mask: In this case, their masks are the same color as their eyelids.
- Expy: Fanboy resembles Sheen from Jimmy Neutron in appearance and behavior. His head also resembles Ed's from Tonic Trouble.
- Fat and Skinny: Chum Chum and Fanboy respectively.
- Fan Boys: So much that they wear costumes that make them look like super heroes for all their life.
- Fanboy even names himself after this trope.
- Fartillery: Chum Chum's underwear "explodes" on Yo in "Prank Master".
- Gasshole: Both of them are this, but it's more prominent with Fanboy.
- Heterosexual Life Partners: To the point where they are intent on becoming "immortal companions" in "Fangboy". In "Separation Anxiety", their friendship is so strong that if they are apart, it causes a rip in the space-time continuum.

Kyle the Conjurer (aka, Kyle Bloodworth Tomison)
- Voiced by: Jamie Kennedy
Kyle is a snarky, grumpy kid who, depending on the episode, takes the role of either the antagonist or a tritagonist for Fanboy and Chum Chum. He actually possesses wizardly powers, although no one believes him.
- Ambiguously Gay: When he and Fanboy are trying to decide which board game to play, his first pick is "Mr. Unicorn's Teddy Bear Dress Up Fashion Show".
- In the "Fanboy and Chum Chum Bible", which is a compliation of ideas Eric Robles wrote down before the show started, Kyle was originally supposed to have a crush on Yo, and although he has never showed any interest in her in the actual series, this could make him Ambiguously Bi.
- When he states that the town isn't big enough for two wizards in There Will Be Shrieks, look at his hand movements.
- In "Fanboy A'hoy!", we get this:

- Tsundere: Type A. He's always rejecting Fanboy and Chum Chum, but it has been hinted several times that he craves their friendship. Especially in the Sigmund the sorcerer episode.
- Vitriolic Best Buds: Fanboy and Chum Chum believe they are best friends with Kyle, even though he makes it clear that he wants nothing to do with them.
- Although, in a show where you wouldn't expect there to be any Character Development, Kyle has actually started liking Fanboy and Chum Chum, and he even admits they're his friends in "Sigmund the Sorcerer."

An incredibly energetic girl who loves her digital pet Yamaguchi. She is one of Fanboy and Chum Chum's schoolmates. Being vulnerable to all things cute and tiny, she becomes extremely obsessive whenever Chum Chum is involved, often to the point of insanity.
- And Call Him George: Yo is obsessed with taking Chum Chum home, dressing him up in a sailor outfit, and putting him on her toy shelf.
- Artificial Limbs: She has a couple of Doc Ock-like robotic arms in her backpack.
- Asian Airhead: Type 2. Though it's not like everyone else is exactly a genius.
- Cell Phone: She has one in "Brain Drain" that allows her to teleport anywhere.

A nerd who lives and runs a comic book shop with his mother. Considered by Fanboy and Chum Chum to be the smartest human being to have ever existed, due to his excessive knowledge of comics.

- Expy He looks remarkably similar to another Comic Book Guy
- Fan Boy: He's more stereotypical example than Fanboy and Chum Chum.

- Companion Cube: He talks to the Chimp Chomp machine as if it's his girlfriend. He's the same way with his car named Sandy.
- High-Pressure Emotion: He has shown this at least three times. In one instance, he gets so angry that the lid to his cup hat blows off.
